I am confused on where to setup talkgroups... is it strictly on the XTS 3000 Radios, Quantar, Centracom or ALL systems?
I have no formal training on Motorola systems so, be detailed if possible.
There is a repeater (duplex freq.) and DIU3000 hooked up to it.
Operator is on a Centracom. I have access to the repeater via RSS and programming software for the XTS3000.
I can see talkgroups in the XTS3000 and change them.

I'm sure no talkgroups are set up in the Quantar... The only thing the Quantar needs is the Network ID (NAC).
Of course you will need to set up your XTS3000s to use talkgroups. Make sure the "Squelch Type" is set to selective squelch so that they only hear their selected talkgroup. Also, I highly recommend enabling "Smart PTT" / "Inhibit on Correct NAC" on any system where multiple talkgroups are used, and this will prevent users that can't hear each other from walking on each other.
The standard DIU only supports transmissions on one talkgroup from a standard tone remote console. The standard DIU does not support "Selective Squelch" or "Smart PTT" like all the mobiles and portables do, so it will hear every talkgroup on the repeater....
The only exception may be if you have the right model of Centracom console that supports the DIU's "Centracom Signalling Link". If your console supports this, you may be able to have the dispatcher hear only their selected talkgroup. And possibly be able to select the talkgroup the dispatcher transmits on.
If you can't set up the "Centracom Signalling Link", you may still be able to get some of the functionality you want if you get a little creative... Adding encryption to everything is a way that you can have the DIU only hear transmissions on a particular talkgroup--just load the correct key for that talkgroup in the DIU. You could then use a different encryption key on talkgroups you didn't want dispatch to hear. The dispatch console will stay muted on those...
Also, be aware that you can program the DIU to transmit on a different talkgroup from the MRTI PATCH connector, and the test handset. I would think that if you interfaced external tone remote panels on the MRTI PATCH and handset connectors, you could transmit on any of the three talkgroups (regular console input, MRTI, or handset) from the tone remote console.
It's really stupid that the DIU doesn't support selectable talkgroup from a standard tone remote, "Selective Squelch", or "Smart PTT". Even Motorola's oldest ASTRO P25 radios supported that stuff. But not the $$$ DIU...
