On the 25 pair Amp connector on the backplane, aren't the PTT, TX and RX audios always available? I mean Tx audio and ground not the balanced audio inputs. The balanced audio inputs are only active with simulcast enabled, are the Tx audio and ground the same? If so, why is the PTT available when simulcast is enabled or disabled? Thanks in advance.

Pretty much everything on that connector requires that the station contain a WLI board.
The same 600 ohm receive and transmit lines that are on the wireline connector are duplicated on connector 17. Use of those as well as PTT & COR is discussed in the "System Applications" section of the manual.
The default PTT in & COR out are actually part of the wildcard programming capability. Master that and you can make the station stand up and do tricks.

XMO is correct about using the J7 50-Pin Telco Connector in conjunction with the station's F9 Wildcard Field. There are two versions of the Wildcard Options X233 Basic or X157 Enhanced. Normally the X233 Basic has the 4-Wire Line board installed into the Quantar / Quantro station.
What the average field service personnel do not realize is, you can take your computer with the correct RSS/CPS interface the programming cable's DB-9 inserted into the station's programming port, READ the station's codeplug, F10 Exit to Main Screen, F2 Service Screen, Enable the Wildcard and then Enable the Wildcard Parameter field for 8-Wire Enhanced although you only have 4-Wire Line I/Os on the module to program what you needed.
If the station had been ordered with the Option X157 Enhanced Wildcard then you would have received the CLN1296A 8-Wire Line I/Os module installed into card cage and been able to program the what you needed. Do not forget to WRITE the Wildcard software programming and the different parameters fields back into the station's codeplug after modifying.

The other thing to know is that, if (as your question implies) you are attempting to use a Quantar with an external repeater controller, you have to remap some of the audio input and output lines, via wildcard programming. There are explitic examples of how to do this in the back of the Quantar maintenance manual.
